What is significance of 25cent coin that has a p rather than RCM identification under the bust of the queen?

It means "plated", as in a thin coating on top of the base metal.

As Bill stated,
It stands for Plated as in Multi-Ply Plated Steel, the new composition for Canadian coins that were previously composed of Nickel. They were now composed of nickel and copper plating over steel.
First introduced in business strikes in 2001, the "P" was used until 2006 when the new RCM logo was introduced.
